In the early phase of apoptosis, the integrity of cell membrane is maintained but the cells lose membrane phospholipid asymmetry. Phosphatidylserine (PS), a negatively charged phospholipid located in the inner leaflet of the plasma membrane, is then exposed at the cell surface. Annexin V, which is a calcium and phospholipid binding protein, binds preferentially to PS. The ANNEXIN V kits are apoptosis detection kits, based on the binding properties of conjugated annexin V to PS, in combination to propidium iodide (PI) staining. Apoptotic cells stained by a simple, quick (10 minutes) one-step staining procedure with the Annexin V-FITC kit. No fixation of the cells is necessary. The stained cells may be analyzed by fluorescence micropscopy or flow cytometry.
