Sharon Soh, Chris Harbert, Francois Degorce, Cisbio International The BioRAPTR FRD Microfluidic Workstation is a platform designed for dispensing low-volume reagents. Its ability to dispense up to eight different reagents simultaneously makes it an ideal platform for the automation of the IP-One HTRF Assay from Cisbio. The BioRAPTR FRD workstation was coupled with the PARADIGM Detection Platform using an HTRF Detection Cartridge for high-throughput automation and detection of the IP-One Assay. The process includes dispensing cells and kit reagents using the BioRAPTR FRD workstation, followed by detection and analysis using the PARADIGM platform. Figure 1. The IP-One HTRF Assay is a homogeneous competitive immunoassay which can be used to monitor Gq-coupled GPCR activity through the inositol phosphate cascade. Inositol Phosphate 1 (IP1) is a downstream metabolite of IP3 which accumulates in cells in the presence of LiCl following Gq-mediated phospholipase C (PLC) activation. For this study, a CHO model system over expressing muscarinic m1 receptor was stimulated with the reference agonists, carbachol and acetylcholine. This poster describes the BioRAPTR FRD workstation, the PARADIGM platform, the automation process and the results obtained using these systems to automate the IP-One assay.
